What BC-700 series reagent set does
The BC-700 series and the BC-6000 go beyond impedance counting and use fluorescence to characterise cells, which is what allows them to report reticulocytes and to flag abnormal populations more confidently than a conventional five-part analyzer can. That capability is why these sets include a fluorescent dye alongside the diluent and the lysing reagents: the dye binds nucleic acid, so cells containing RNA or DNA fluoresce and can be separated from those that do not. It is the reagent that makes reticulocyte counting possible on an automated platform, and it has no equivalent in the simpler sets, which is one clear reason these fluids cannot be substituted.

These are the tertiary hospital and referral laboratory instruments, so this set goes to a smaller number of higher-volume sites than the BC-5000 series does, and those sites usually have the tightest turnaround expectations and the most formal quality requirements. The dye is the item most often under-ordered, because it is consumed in smaller volumes than diluent and is easy to overlook against the bulk items, but running out of it costs the reticulocyte count and the extended differential rather than the whole CBC. Storage and handling
Store containers upright and sealed until placed on the analyzer, within the labelled temperature range. The fluorescent dye is light-sensitive and has its own storage requirements distinct from the diluent and lyse, so it should not be stored simply alongside them. What is the fluorescent dye for?
It binds nucleic acid, so cells containing RNA or DNA fluoresce and can be separated from those that do not. That is what makes automated reticulocyte counting possible, since reticulocytes are young red cells that still contain RNA. It also helps the analyzer flag abnormal populations. No simpler reagent set contains an equivalent.
What happens if we run out of dye but still have diluent and lyse?
The analyzer will still produce a complete blood count, but you lose the reticulocyte count and the extended flagging that the fluorescence channel provides. Because the basic count keeps working, this is a stock-out that can go unnoticed until a clinician asks why reticulocytes are no longer being reported, which is why the dye should be tracked as part of the set.
