How chemiluminescence immunoassay works

A chemiluminescence immunoassay measures light rather than colour. The reagent carries antibodies bound to microscopic paramagnetic particles, and those antibodies capture the one analyte being measured out of the patient sample. A second antibody carrying a chemiluminescent label binds to the captured analyte as well, so each captured molecule ends up sandwiched between a particle and a label. A magnet then holds the particles in place while everything unbound is washed away, and that step is what gives immunoassay its sensitivity: what remains is only what was genuinely captured. Adding a trigger solution makes the label emit light, and the analyzer counts photons. More analyte means more label retained and more light. That is why wash buffer and substrate solution are consumables in their own right rather than accessories, and why they are ordered alongside every assay on the menu. For very small analytes a competitive format is used instead, in which the patient sample and a labelled version of the analyte compete for a limited number of antibody sites, so the light signal falls as concentration rises.
