What Estradiol measures
Estradiol is the most active of the oestrogens, produced mainly by the developing ovarian follicle, and its concentration rises and falls in a characteristic pattern across the menstrual cycle. It is measured in the assessment of infertility, in the investigation of absent or irregular periods, in the assessment of ovarian function, and in monitoring during assisted reproduction, where serial measurements track the response to stimulation. As with FSH, a single value carries meaning only alongside the day of the cycle it was taken on, and in a monitoring context it is the sequence of values rather than any one of them that guides what happens next.

Why is estradiol measured repeatedly during a fertility cycle?
Because it tracks the developing follicles, so the sequence of values shows how the ovary is responding to stimulation and allows treatment to be adjusted while the cycle is still running. A single measurement in that setting would be far less useful than the series, which is why fertility monitoring consumes considerably more reagent per patient than a diagnostic request does.
